I give you hints.

There's something about the skeleton. If you did crimson room, think along the line of the curtain in crimson room. The locked book plus the blanket has a lot of connections to the skeleton.

The characters are used to make something. But in a specific manner. You need to find out the manner and you need the item to start making the something.

If you took all your items, and all the spaces in the inventory column is filled means you didn't miss out anything.

Read "The Locked Book" again. There are clues inside what to do. Think along the line of "Cold".

You need of course a key. But it's not easy to find that one.

Hint:
A lot to do with the skeleton
If you played Crimson room, think along the lines of the curtain.
If you didn't play Crimson room, think along the line of the paper with the hair
Very related to the medicine bottle
